
Even though there are many sources of molecular biology protocols including popular published laboratory manuals such as Sambrook et al. and others, there is a need to adopt these protocols to the particular circumstance in your own lab. In our lab, we have optimized various protocols taking into consideration the cost, efficiency, and suitability for the lab environment. Particularly, the DNA extraction protocol we have (a modificiation of Dellaporta et al.) is very robust and applies to various crops.
Protocols for DNA quantification and gel electrophoresis were included for educational purpose only. These are established methods that entails little modification. Likewise, genotyping with silver staining (PAGE) for SSR and AFLP are also based on manufacturer's recommendations of the kit we use.
